I'm not quite sure whether this should be part of vehicle or route construction... is it possible to edit destination boards (ad different destinations) to existing buses/routes?

Also, is it possible to edit timetables on existing route?

Yes it is. Chapter 5 in the Manual (right now only aviable in german)
